چکیده

An enzyme which catalyzes the conversion of L-tryptophan to indole-3-acetamide has been isolated from cells of Pseudomonas savastanoi and purified 89-fold. The enzyme catalyzes a reaction in which 1 mole of L-tryptophan and 1 mole of 02 are consumed and 1 mole of indole-3-acetamide and 1 mole of CO2 are produced. The enzyme has a pH optimum between 7.2 and 9.2. It is specific for the L isomer of tryptophan and attacks 5-hydroxytryptophan at a rate approximately 17% of that for L-tryptophan. It shows slight activity toward L-phenylalanine and L-tyrosine but no activity toward indole-3-pyruvic acid, indole-3-acetaldehyde, amethyltryptophan, or indole-3-acetic acid. Indole-3-acetamide brings about end product inhibition and, at a concentration of 1 mM, reduces the rate of oxidation of L-tryptophan by 85 %. Hydroxylamine, HgCI 2, iodoacetamide, and indole3-acetic acid are effective inhibitors of the enzyme. Tryptophan oxidative decarboxylase is suggested for the trivial name for this enzyme.

Indole-3-acetic acid (IAA) synthesis in the biocontrol strain CHA0 of Pseudomonas fluorescens: role of tryptophan side chain oxidase.

Pseudomonas fluorescens strain CHA0 is an effective biocontrol agent against soil-borne fungal plant pathogens. In this study, indole-3-acetic acid (IAA) biosynthesis in strain CHA0 was investigated. Two key enzyme activities were found to be involved: tryptophan side chain oxidase (TSO) and tryptophan transaminase. TSO was induced in the stationary growth phase. By fractionation of a cell extr...

Bioproduction of Indole Acetic Acid by Rhizobium Strains Isolated from Root Nodules of Green Manure Crop, Sesbania sesban (L.) Merr.

Twenty six Rhizobium strains were isolated from root nodules of Sesbania sesban (L.) Merr. collected from different regions of Andhra Pradesh. All the 26 Rhizobium strains produced indole acetic acid (IAA), but maximum amount was produced by only five strains in yeast extract mannitol  (YEM) medium supplemented with L-tryptophan. The strains were found to elaborate maximum IAA when fed with 2.5...

Auxin production is a common feature of most pathovars of Pseudomonas syringae.

We investigated indole-3-acetic acid (IAA) production by 57 pathovars of Pseudomonas syringae and related species. Most of those analyzed produced IAA, especially in the presence of tryptophan. Eight strains produced high IAA concentrations in the absence of Trp. The iaaM and iaaH genes of P. savastanoi pv. savastanoi were detected in a limited number of strains only, including the eight above-...

Indole-3-acetic acid biosynthesis in colletotrichum gloeosporioides f. sp. aeschynomene

We characterized the biosynthesis of indole-3-acetic acid by the mycoherbicide Colletotrichum gloeosporioides f. sp. aeschynomene. Auxin production was tryptophan dependent. Compounds from the indole-3-acetamide and indole-3-pyruvic acid pathways were detected in culture filtrates. Feeding experiments and in vitro assay confirmed the presence of both pathways. Indole-3-acetamide was the major p...

IAA produced by PGPR probably plays an essential role in the plant-microbe interactions, However, regulatory mechanisms for IAA production in the bacteria have not yet been identified. In t...
